If you have the patient’s absolute lymphocyte count (ALC), call 513-636-4685 or use the "Customized Volumes by ALC" sheet to obtain the precise amount of blood required for this assay. If you do not have an ALC available, 5-10mL blood in Sodium Heparin (Dark Green) is the general requirement and should be sufficient for patients who are not lymphopenic.

Peripheral blood lymphocytes that have previously been exposed in vivo to antigens become sensitized. If they are subsequently exposed in vitro to those same antigens, they will be triggered to initiate an immune response that culminates in proliferation. This assay uses the antigens Candida Albicans and tetanus toxoid to stimulate a purified mononuclear cell preparation and test for the ability of the cells to respond by proliferating. An excess of radio-labeled thymidine is made available to the cells during the later part of the culture period. Cells undergoing DNA synthesis will incorporate the thymidine, which can then be measured using a microplate scintillation counter.
