Real-time PCR

Use

Rapid diagnosis of infections caused by C. trachomatis (e.g., cervicitis, PID, urethritis, nongonococcal urethritis, and sexually transmitted diseases) and N. gonorrhoeae. The sensitivity of the test exceeds that of culture and other nonmolecular assays.

Limitations

This test is not approved for conjunctival and most respiratory specimens, nor can it be used as a test of cure or in cases of suspected sexual abuse. A negative result does not exclude infection.

Additional Information

When a specimen with a positive C. trachomatis and/or Neisseria gonorrhoeae is resulted, LA County Public Health Morbidity Unit requires the completion of a STD Confidential Morbidity Report by both the performing laboratory and the Health Care Provider. Collection Instructions

Urine: Collect the first 20-30 mL of voided urine from patients who have not urinated within 1 hour prior to collection. Collect and transport in a sterile urine container.
Endocervical swab: Use the Universal Viral Transport Kit for specimen collection and transport. Remove excess cervical mucus from endocervix before collecting specimen. Insert the swab into the endocervix to sample columnar and squamocolumnar cells and rotate for 15-30 seconds. Place in transport kit.
Urethral swab: Insert the mini tip flocked swab 2 to 4 cm into the urethra and rotate for 2 to 3 seconds. Place swab in Universal Viral Transport.
Note: Do not use swabs on wooden or aluminum shafts, these interfere with assay.

Volume

Collect 20-30 mL of first voided urine. One endocervical, urethral, vaginal, anal/rectal, or pharyngeal swab in Universal Viral Transport.

10 mL first voided urine, one swab in Universal Viral Transport, 3 mL Cytyc PreservCyt® (ThinPrep®) or SurePath® liquid cytology media
