QuantiFERON® - TB Gold PLUS Collection Video

4 special QuantiFERON® - TB Gold PLUS high altitude (HA) collection tubes

Required volume: 1.0 mL blood (red arrow) per tube

Please call lab to request tubes, 720-848-4401
  1. Lawson #312100, QFT-Plus HA Collection tubes
  2. Collect 1 mL of blood into each of the 4 QuantiFERON Gold PLUS collection tubes for High Altitude (HA).
  3. Tubes do not have to be drawn in any particular order. Keep tube and needle in place for 2-3 seconds after tube appears to have completed filling and ensure correct volume collection.
  4. Tubes have low vacuum and fill extremely slowly.
  5. To avoid underfilling tubes: it may be helpful to collect patients’ blood sample using a syringe. With the full syringe inoculate the 4 tubes to their fill lines. Do not allow blood to sit in syringe, fill 4 tubes immediately.
  6. Immediately after collection, shake tubes 10 times firmly to ensure the entire inner surfaces of the tubes are coated with blood to dissolve the antigens on the tube walls.
  7. Avoid overly vigorous shaking, this may cause gel disruption and aberrant results.
  8. Label each of the four tubes and include the time of collection.
  9. Maintain and transport tubes at ambient temperature. Time sensitive.
Acceptable sample:
Blood volume fill level in each tube is within the black bar.
All 4 tubes must be submitted within 16 hours of collection at room temperature.

Unacceptable sample:
Volume of blood collected is above or below the black bar on the label.

Specimen Preparation

UCHealth labs prior to sending samples MUST prepare samples following instructions below:
    1. At the local clinical lab, prepare samples for plasma separation following:
      1. The 4 collection tubes may remain at room temperature up to 16 hours from time of collection.
      2. Examine all 4 tubes for proper fill level
      3. If any of the 4 tubes is over or under acceptable fill level, cancel test and notify unit for recollection.
      4. When all 4 tubes have acceptable blood volume, continue.
    2. Incubate tubes upright at 37°C for 16-24 hours.
    3. After incubation, centrifuge tubes @ 4600-5000 rpm or 2000-3000 rfc for 15 minutes, clear plasma separation
    4. Once tubes are spun, refrigerate tubes (stable 28 days). Samples are now ready to transport to UCH lab for testing.
  1. Ready Samples for Transport:
    1. Create packing list and SCAN ALL 4 tubes to packing list and send refrigerated.
    2. Bag each patient's 4 tubes in one Biohazard bag, or if multiple patients, place samples in a rack and cover with bag.
    3. Transport tubes refrigerated to UCH/MetroD lab for testing.
    4. Send one shipment a day to UCH, preferably in the morning.

Unacceptable Conditions

  1. Tubes that are over- or underfilled: If volume of blood collected is above or below the black bar indicated in photo, the sample is unacceptable.
  2. Incorrect collection tubes.
  3. Unspun whole blood refrigerated or frozen samples.
  4. Specimens not properly identified.

Storage/Transport Temperature

Internal: Deliver to lab immediately at ambient temperature before 8:00 pm Monday-Friday, same day of collection. Time sensitive.
Offsite: Obtain collection containers from UCH Clinical lab, collect samples as indicated and return to UCH Clinical lab the same day of collection before 8:00pm Monday-Friday, Time sensitive.

UCHealth labs MUST prepared samples as stated above in SPECIMEN PREPARATION before transporting.

Ordering Recommendations

A quantiferon should not be used alone to rule out active tuberculosis. It is negative in 20% of cases of active Tuberculosis. Sputum cultures and/or tissue samples are needed to rule out active tuberculosis.

Interpretive Data

A negative result must be correlated with the individual’s medical and historical data, particularly for individuals with impaired immune function:
  • HIV infection or AIDS
  • Treatment with immunosuppressive drugs
  • Hematologic disorders (e.g. leukemias, lymphomas, myeloproliferative   disorders)
  • Specific malignancies (e.g. carcinoma of the head, neck, or lung)
