Collect

Preferred: 2-5 mL of aseptically aspirated body fluid in a sterile capped syringe or sterile container. Nephrostomy urine may be submitted in a sterile container or Grey Top.
Acceptable: Peritoneal or synovial fluid in blood culture bottles are accepted, but not preferred. To perform a stain fluid must also be submitted in a sterile container.
Minimum volume: 1 mL in a sterile container (contact lab if less).

Body fluid examples:
  • Ascites/peritoneal/paracentesis fluid
  • Synovial/joint/bursa fluid
  • Pericardial fluid
  • Pleural/thoracentesis fluid
  • Nephrostomy urine (for other urines: see LAB239 Urine Culture).

Unacceptable Conditions

  • Medical devices.
  • Urinary catheters.
  • Swabs.
  • Body fluid submitted in preservative or media (e.g., EDTA, heparin, formalin, etc.).
  • Grossly contaminated specimens.
  • Sample not properly identified, incorrect container, insufficient volume, or past stability.

Storage/Transport Temperature

Internal: ASAP* ambient
Offsite: Transport ASAP.* Ambient preferred.

*Delay in transport may result in overgrowth of contaminating flora.

Refrigerated: Refrigeration may compromise the recovery of fastidious organisms
Frozen: Unacceptable

Remarks

  • This test is for anaerobic culture of body fluids only (includes gram stain; sensitivity testing will be added when needed).
    • For body fluid aerobic, order LAB269 Body Fluid Culture, Aerobic.
    • For CSF anaerobic, order LAB3354 CSF Anaerobic Culture.
    • For BAL or other respiratory sources, order LAB3277 Respiratory Culture or LAB3696 Cystic Fibrosis Respiratory Culture.
  • Provider is notified of all positive smears or cultures. Negative cultures are reported at 48 hours with broth held for 7 days prior to discarding.

Anaerobic culture

Organism(s) recovered in culture may be identified by MALDI-TOF. Identification by MALDI-TOF was developed and its performance characteristics determined by the University of Colorado Hospital Authority.  It has not been cleared or approved by the U.S. Food and Drug Administration.  The laboratory is regulated under CLIA as qualified to perform high-complexity testing.  This test is used for clinical purposes.  It should not be regarded as investigational or for research.
